Two words: female intuition.
Females are very perceptive when it comes to these things -- if you think he likes your friend, he probably does.
You don't say how well you know this guy -- is he a friend too?
If yes, you can just keep being his friend & ask him if he likes your friend. If he does, don't dismay - if may turn out that later on, the more your friendship develops, he'll come to like you.
If no, get to know him. Be casual - just smile nicely when you see him, throw in the occasional 'hi' as you go by... you want him to feel comfortable around you. When he does, you can either become his friend (as per above) OR he might start noticing you instead of your friend. Just don't throw yourself at him if he does start to show some interest at you -- that's a sure-fire way to lose him!

First of all, try not to assume too quickly about who he likes. But if you strongly think that, why not ask him in person?
I know, it's easier said than done.
Ask your best friend how she feels about this as well, does she know about it how you feel for him or how he feels for her?
The best thing NOT to do is let any jealousy get in the way, try and be calm and composed about this as best as possible.
In any case, the best thing you can do right now is talk to your best friend about this first. Write back to me if you want with what happened when you've done that.
