Your question dosent give much to go on.

Think about your friendship with your best friend, will he be upset with you? Will he be protective of his sister? Will it damage your friendship?

Think about what means more, a girl giving mixed signals or your friendship? I beleive that asking yourself this will solve your problem.

women do like playing with mens minds more so if they sure you like them. you have to ask her one day if she would like to go on a date. the worst that can happen is she will say no and you just say cool no harm trying. if your friend is a true friend you can explain to him afterwards that you have spent alot of time growing up with his sister through him and you have grown to admire her and would like it very much to take her for a meal thats all. but keep it light so no one feels to uncomfortable. girls are flattered when men like them, and if nothing else you will be helping perfect your skills for asking women out and atleast its someone you know, you can ask her advice if she says no, on the best ways to chat to women, become her friend and she may end up liking you that way. any good women is worth waiting for but she must know your willing to put yourself on the line for her. good luck
