He has not told me or anything but I just have this feeling because lately he is acting more mature around me and a couple weeks ago he put his arm around me at a football game. I am afraid to tell my best friend what I think because I know she would not be ok with us dating, so do I tell her anyways? or do I keep it a secret if something were to happen between us? I really dont want to ruin our friendship.

Definitely don't keep it a secret because your friend will find out eventually and it will hurt her more if you kept it hidden from her.
You don't need to tell her anything until something happens for certain. There's no need to cause unnecessary hurt if nothing is going to happen anyway.
If you and this guy start dating or whatever you do need to sit your friend down and explain to her how you feel about her brother. Just be sure this is what you really want because if you and this guy don't work out you might be ruining a friendship too.
