Honestly, I find the act of 'hitting' on a girl annoying in itself. Girls can generally tell when you're just being friendly, and when you've got an agenda. But then, that may just be me.
Body language is key. There are all kinds of messages you can get from the way a lady carries herself. Some of the 'go' signals can include turning toward you when you speak to her, lots of eye contact, smiling, playing with her hair, hands at her side or on her hips.
Some of the 'go-away' signals are things like keeping her back or side to you, looking around or doing other things when you speak to her, no eye contact, one-word answers and crossed arms.
You've also got to use your common sense. If she's looking busy, harassed, or distracted, then you're probably out of luck.
