I'm a theatre major, been doing it for years and in that time I've done so many stage kisses it's ridiculous (and borderline slutty!).
The only way you'll "mess it up" is if you freeze up, stop moving completely (don't be a statue!) and don't kiss him back. You've seen plenty of movies, I'm sure... I want you to go rent on with steamy kisses (and chick flick will do) and I want you to LITERALLY watch and learn. Remember to touch his face or wrap your arms around him.
It'll be over as quickly as it began. You say that Tony is a good friend of yours. Does he know it's your first kiss? He's the guy, he should be taking control anyways. Tell him that you've never really kissed a guy before and he should show you what to do. Practice privately - it's a whole lot less embarrassing than doing it in front of your director and cast!
Just remember that it's not YOU kissing, it's Maria. Don't be nervous about any romantic stuff, just make it look good for the audience and you'll be fine.
