Talk to him and explain how much you like her... If he's a good friend, you'll understand... Unless your best friend is a girl, which in that case, is a hell no, choose your best friend...

I guess ultimately the question comes down to which you value more: this friendship or the possibility of a romantic relationship with your friend's sister (and really, it's going to be hard if not impossible to have both). Do what you have to do, but there are a lot of girls out there, and do you know whether your friend's sister is even interested in you? If I were you I'd leave the sister alone.
