For example, today I was sitting at a table opposite a friend at work. This girl who I vaguely knew sat down next to me, but kind of kept her gaze on my friend who sitting across from us. When I got up to leave, she was really softly spoken and helped move her chair for me to get out.Does this mean a girl totally does not have a crush and doesn't want to attract your attention? Or does it mean she does have a crush, and is shy or unsure of me?Thanks.

She's paying attention to whoever she finds the most attractive. This does not mean she has a crush, that that she's attracted - there's a difference. It also does not mean you have no shot. One of the downsides of being a guy is you're going to have to do the approaching, often without any indicator that she's even interested until later.
There is nothing wrong with hitting on a girl only to find out she's not attracted. By trying to anticipate her interest before you've even attempted to create it, you're missing a lot of great opportunities. Flirt a little more freely, they might be attracted to the boldness of it.
