Depends on your culture.
It can mean the moment that the Hymen (a layer of skin across the vagina) is broken. But since that now happens mostly by accident over a period of years through cycling, sports, dancing, tampons, running etc, that doesn't hold much meaning.
Most girls take it to mean the first time they have sex with a man.

"Losing your virginity" refers to that first time you have sexual intercourse with another person. It's more a social term that determines you've had a close sexual relationship for the first time rather than a scientific term suggesting you have been penetrated, whether it by a person or an object. The word "virgin" is usually applied to mean "innocent" or "untouched" and this is "taken away" by another person.
